The Courtyard Key is located in the Wine Room in Castle Dimitrescu, but you'll need to put in a bit of work to actually obtain it. The sequence you need to trigger in order to start the path to getting the Courtyard Key begins after placing the Maroon Eye in the Prioress Door on the second floor of the Main Hall. One of Lady Dimitrescu's daughters will then spawn and chase you through a series of new rooms and areas, including a dark jail. This will eventually turn into a boss fight in which you must deal damage to her when she is standing in front of the open window.

Once she's dead, grab the Sanguis Virginis wine bottle from the large bowl of blood in the Kitchen and then make your way back to the Wine Room by following the new path you've opened up. Place the Sanguis Virginis on the pedestal in the Wine Room and a new area will appear. The Courtyard Key is inside, contained in a small treasure box.
