In the Downtown chapter, you'll end up in a subway, around the time you collect a Molotov for the first time. You'll reach a waiting room area with a selection of stores, which is inhabited by Clickers and Runners. We'd recommend you deal with these first, before proceeding with how to open the Safe in Downtown.

Once the area is clear, go inside the medicine shop on the left of the waiting room, and collect the Artefact named Note to Derek. This has the Safe code combination written on it, which is 03-43-78. Now, on the opposite side of the room, you'll find a shop called Round Note. Underneath the counter, is the Downtown Safe location, where you can input the code.
